Study Overview
In 2003, Dr. Walter Canonica and his team at the University of Genoa, Italy, conducted a landmark study on immunotherapy for allergies. This study remains one of the most significant contributions to the field in part because it contains the first long term, longitudinal study proving continuing effectiveness.
Dr. Canonica and his team observed that various modalities of immunotherapy have been utilized for over a century, starting with subcutaneous immunotherapy (SCIT). They explored different methods including oral, bronchial, and nasal immunotherapy, ultimately concluding that sublingual immunotherapy (SLIT) offers the best balance of efficacy and safety.

Immunotherapy modalities have evolved over the past century, with SLIT emerging in the mid-1980s.
Effectiveness and Safety:
- A “double-blind double-dummy” study demonstrated equivalent effectiveness of SLIT and SCIT.
- Another study found that SLIT had far superior safety and tolerability compared to SCIT.
- Pediatric studies indicated negligible side effects for SLIT, with only 3% of patients experiencing side effects.
- SLIT controlled symptoms sooner than SCIT.
Compliance and Convenience:
- SCIT discontinuation rates ranged from 10% to 34%, often due to the inconvenience of clinic visits.
- SLIT allows for at-home treatment, significantly improving patient compliance and adherence to the treatment regimen.
Long-term Benefits
A ten-year longitudinal study demonstrated that patients who completed SLIT treatment for allergic asthma remained symptom-free ten years later.
